Butyrate
Also sold as: Sodium butyrate · Butyric acid · Tributyrin
A short-chain fatty acid produced by microbial fiber fermentation and sold as salts or tributyrin. Oral supplement evidence is much narrower than the established biological role of endogenous butyrate.

Research
- Focus: No focus-enhancement RCT is supplied. Depression associations and gut-brain mechanisms cannot establish cognitive efficacy; no negative focus RCT is implied.
REALITY CHECK // Butyrate is not a proven SIBO or depression treatment. Oral IBS findings are small and often concern mixtures, while better-documented enema studies do not show consistent UC benefit. Preclinical and biomarker effects should not be presented as clinical cures.
Levels: how we grade evidence. Grey = evidence of no benefit; amber = evidence it makes this goal worse.
Ratings are self-reported by a self-selected group of users: how it felt, not proof that it works.

How it was studied
UPPER LIMIT // No EFSA/NIH tolerable upper intake level is established. Research around2 g/day and pediatric weight-based trials are not universal safety ceilings; long-term safety is incompletely characterized.
Summary
IBS findings are preliminary and often confounded by probiotics/prebiotics. A review of8 enema RCTs in227 UC patients did not support clinical use, and a72-patient pediatric oral IBD trial was null. Small metabolic/weight studies are promising but insufficient for a general gut or metabolic cure.
Mechanism
Butyrate fuels colonocytes and influences barrier signaling and histone deacetylases. Endogenous fermentation evidence does not prove that an oral supplement adds benefit beyond adequate fiber; gut use and first-pass metabolism limit systemic exposure.
Typical studied dose
The pediatric IBD trial used150 mg sodium butyrate twice daily for12 weeks and was clinically null. A46-adult proof-of-concept trial used1875 mg/day for12 weeks. Pediatric obesity research used20 mg/kg/day for6 months under supervision; no pediatric self-dosing protocol is implied.
Upper limit
No EFSA/NIH tolerable upper intake level is established. Research around2 g/day and pediatric weight-based trials are not universal safety ceilings; long-term safety is incompletely characterized.

Forms and label traps
| Form on label | Active content | Studied range / day | Absorption & GI tolerance | Verdict |
|---|---|---|---|---|
| Sodium butyrate Best for: Identify the exact salt mass in the adult proof-of-concept context. |
Extract weight · ~100% marker | 1875–1875 mg | Rapid upper-GI uptake and metabolism can limit colonic delivery. Strong odor; higher amounts may cause gas or abdominal discomfort. |
Studied standard [FORM // TRIAL_CONTEXT] 1875 mg/day is one12-week trial amount, not a general target;100% here means declared sodium-butyrate ingredient mass, not elemental butyrate or sodium. |
| Microencapsulated sodium butyrate Best for: Match capsule coating, active salt content and indication. |
Extract weight · ~100% marker | 300–300 mg | Coating modifies release and masks odor; superior clinical outcomes are not established. Often tolerated; the pediatric IBD trial reported no adverse events. |
Good option [FORM // TRIAL_CONTEXT] 300 mg/day belongs to a supervised pediatric IBD trial with no added clinical benefit, not an effective adult IBS dose. |
| Tributyrin precursor Best for: Sodium-free precursor comparison. |
Unstandardized: active content unknown | no comparable trial range | Release rationale includes in vitro digestion and animal colitis research. Digestive tolerance varies; stop for persistent symptoms. |
Caution [FORM // NO_VALIDATED_RANGE] No verified human dose interval or head-to-head clinical superiority is supplied. Do not equate precursor mass with delivered colonic butyrate. |
| Calcium/magnesium butyrate salts Best for: Compare the exact preparation and declared ingredient mass. |
Unstandardized: active content unknown | no comparable trial range | No reliable human head-to-head superiority established. Digestive tolerance varies; stop for persistent symptoms. |
Caution [FORM // NO_VALIDATED_RANGE] No human head-to-head advantage or verified active fraction supplied. Mineral content must come from the actual label. |
Common label traps
- TRAP_01 // CURES SIBO
No SIBO-treatment trial is indexed in this dossier; absence of evidence is not a negative RCT. - TRAP_02 // COATING OR TRIBUTYRIN GUARANTEES SUPERIOR RESULTS
Delivery rationale is not a demonstrated clinical head-to-head benefit. - TRAP_03 // GUT-BRAIN MECHANISM MEANS ANTIDEPRESSANT
SCFA associations may reflect reverse causation; no human oral-butyrate depression-treatment RCT is supplied.

PMID 36014789 PMID 32476236 - IBS SYMPTOMS Limited · benefit
Mixture RCTs and an open/cohort study suggest symptom relief; a colonoscopy model demonstrates an acute permeability effect rather than durable cure.
PMID 39797089 PMID 35371361 PMID 40810534 PMID 40575487 - METABOLISM / WEIGHT Limited · benefit
A46-adult12-week study at1875 mg/day reported weight change in participants without diabetes of-7.0±3.0 versus-3.2±1.6 kg (p=0.001); T2D triglycerides changed-0.36 versus+0.08 mmol/L (p=0.012), with+9% time in tight range. These small proof-of-concept findings require replication.
PMID 41880673 - PEDIATRIC OBESITY Limited · benefit
A6-month20 mg/kg/day trial favored BMI-SDS improvement; two published corrections mean the exact effect magnitude needs checking. No pediatric protocol is inferred.
PMID 36469320 - LIVER DISEASE Limited · benefit
Human findings are preliminary and subgroup-based; the positive NAFLD meta-analysis is preclinical, not human efficacy evidence.

Reader questions
Is sodium or calcium/magnesium butyrate better?
The minerals are counterions. Consider actual mineral exposure and release system; no reliable clinical head-to-head superiority is supplied.
Do gas and insomnia mean I should push through?
No. Gas is a reported tolerability issue and insomnia reports are anecdotal, not a known required adaptation. Stop for marked or persistent symptoms and review coingredients.
Is tributyrin worth a premium?
Its delivery rationale has in vitro/preclinical support, but this dossier cannot establish superior human outcomes or cost-effectiveness.
Can I use it for a toddler with suspected SIBO?
No home protocol follows from these data. Persistent symptoms in a child require pediatric assessment; supervised IBD/obesity trials do not establish SIBO efficacy.
Stack interactions
- Caution Glucose-lowering drugs [CAUTION // CLINICAL_CONTEXT]
A42-patient6-week T2D study had nonsignificant between-group glycemic findings; monitor clinically if adding a supplement, without assuming a reliable drug-like effect.
PMID 35623879 PMID 41880673 - Caution Antihypertensives [CAUTION // CLINICAL_CONTEXT]
Within-group BP falls in a small T2D study do not establish a placebo-adjusted antihypertensive action; any additivity remains theoretical.
PMID 35623879 - Caution FOS, inulin, PHGG and probiotics [CAUTION // CLINICAL_CONTEXT]
Combination IBS studies cannot separate butyrate from coingredients. Fermentable fiber can increase gas or bloating in fermentation-sensitive people.
PMID 39797089
Red flags: interactions and who should avoid it
Interactions
Possible glucose/BP additivity is theoretical and based on small inconsistent trials. Probiotics and fermentable fibers often accompany butyrate, obscuring its independent benefit; fibers can aggravate gas in susceptible patients.
Avoid or ask a doctor first if
Pregnancy/breastfeeding safety at supplemental doses is unestablished. Pediatric trials were supervised. Persistent pain, severe digestive symptoms or marked insomnia warrant stopping and assessment; sodium-containing forms matter when sodium restriction is required.
