Research

  • Sleep: No indexed sleep/stress/fatigue RCT is supplied here; absence of indexed evidence is not a negative sleep trial.
  • Focus: Small often industry-associated trials report memory/attention improvements after 6-12 weeks; independent replication and clinical importance remain uncertain.
  • Energy: A 23-man exercise RCT found no aerobic-performance advantage despite a mitochondrial-marker increase.

REALITY CHECK // A measurable mitochondrial marker is not an endurance benefit: the exercise RCT was functionally negative. Cognitive findings are promising but small and often industry-linked. Immediate energy, sleep improvement, proven CoQ10 synergy and human mitochondrial-density claims outrun this evidence.

Levels: how we grade evidence. Grey = evidence of no benefit; amber = evidence it makes this goal worse.

Ratings are self-reported by a self-selected group of users: how it felt, not proof that it works.

How it was studied

01 // STUDIED_DOSE_RANGE
Trial quantities, not a prescription
PQQ disodium studies generally used 10-20 mg/day; a cognition trial used 20 mg/day for 12 weeks and an exercise RCT used 20 mg/day for 6 weeks. A 10-person metabolic study used 0.3 mg/kg/day for about 76 hours. A reduced/dihydro-PQQ MCI trial lasted 6 weeks, but its exact dose is not supplied here. Do not infer 50-100 mg regimens.
02 // TIMING_&_MATRIX
Preparation and administration
No optimized fed/fasted or morning/evening schedule is established by the supplied trials. Morning use is a practical response to anecdotal stimulation, not a proven efficacy requirement. Match the actual named salt/form.
03 // EVALUATION_WINDOW
Match the study window
Cognitive trials ran 6-12 weeks; the exercise trial ran 6 weeks. The 76-hour metabolic experiment is a small biomarker study and does not predict chronic symptom relief.
04 // SAFETY_&_QUALITY_GATE
Check eligibility before use
Require identity/purity testing and an explicit PQQ amount. Do not extrapolate a reduced-form trial to a different salt/dose, or turn a supplement into a fertility or anticancer protocol.

UPPER LIMIT // No EFSA/NIH UL is documented in the dossier. Ten to twenty mg/day is a studied range, not a safety ceiling; long-term safety beyond approximately 12 weeks is insufficiently characterized. Regulatory/novel-food eligibility is a separate question requiring current verification before commercial claims.

Summary

Several small cognitive RCTs report selected improvements, commonly with 20 mg/day for 12 weeks. Trials are often manufacturer-associated and lack large independent replications. In 23 untrained men, 20 mg/day for 6 weeks with endurance training increased a mitochondrial signaling marker without improving aerobic performance. PQQ has no demonstrated human longevity or immediate energy-rush benefit.

Mechanism

Cell/animal studies link PQQ to CREB/PGC-1alpha, SIRT1 and AMPK mitochondrial signaling and redox cycling. Human metabolite or signaling-marker changes do not establish increased functional mitochondrial capacity or clinically meaningful energy.

Typical studied dose

PQQ disodium studies generally used 10-20 mg/day; a cognition trial used 20 mg/day for 12 weeks and an exercise RCT used 20 mg/day for 6 weeks. A 10-person metabolic study used 0.3 mg/kg/day for about 76 hours. A reduced/dihydro-PQQ MCI trial lasted 6 weeks, but its exact dose is not supplied here. Do not infer 50-100 mg regimens.

Upper limit

No EFSA/NIH UL is documented in the dossier. Ten to twenty mg/day is a studied range, not a safety ceiling; long-term safety beyond approximately 12 weeks is insufficiently characterized. Regulatory/novel-food eligibility is a separate question requiring current verification before commercial claims.

Forms and label traps

Forms compared
Form on labelActive contentStudied range / dayAbsorption & GI toleranceVerdict
PQQ disodium salt, branded or generic
Best for: Matching the main salt used in the small cognitive/metabolic trials.
Extract weight · ~100% marker 10–20 mg Water-soluble named salt with human trial exposure data; branding alone does not establish greater potency.
Generally tolerated in short 10-20 mg/day studies; combination-product GI reports cannot isolate PQQ.
Studied standard [REFERENCE // CHECK_PREPARATION]
Compare declared PQQ disodium compound mass, not elemental sodium or an absorption-scaled amount.
Reduced/dihydro-PQQ (PQQH2)
Best for: Identifying the exact reduced preparation used in the 6-week MCI study.
Unstandardized: active content unknown no comparable trial range Reduced-form chemistry and a small MCI RCT do not establish superiority over disodium PQQ.
No strong comparative tolerability data are supplied.
Caution [CAUTION // CHECK_PREPARATION]
Exact trial dose is unavailable here; no assumed mg equivalence or bioavailability multiplier.
PQQ free acid
Best for: Recognizing an uncommon preparation with limited evidence transfer.
Unstandardized: active content unknown no comparable trial range Less soluble than the disodium salt in the dossier; clinical comparative data are absent.
Preparation-specific GI data are unavailable.
Caution [CAUTION // CHECK_PREPARATION]
No supplied exact human range or conversion from free acid to trial salt; efficacy comparison unavailable.

Common label traps

  • TRAP_01 // Instant mitochondrial energy rush
Anecdotal sensations do not establish efficacy. The human endurance RCT found no functional performance improvement.
  • TRAP_02 // Builds more mitochondria, therefore better performance
PGC-1alpha and metabolite changes are biomarkers; the exercise RCT illustrates that function may remain unchanged.
  • TRAP_03 // Branded or reduced PQQ is proven stronger
The same named salt does not become a different molecule through branding, and reduced-form superiority lacks human head-to-head evidence.
  • TRAP_04 // PQQ plus CoQ10 has proven synergy
A mechanistic pairing and popular stack are not a clinical synergy trial.
  • TRAP_05 // High-dose PQQ is a fertility or cancer treatment
Preclinical mechanisms and uncontrolled AMH findings do not establish clinical efficacy or safe high-dose treatment.

Other outcomes studied

Outside the five leaderboard goals. Same evidence levels as above.

  • Memory and attention Limited · benefit
Small trials report selected cognitive improvements: 20 mg/day for 12 weeks in adults aged 20-65, and 6-week reduced-PQQ data in older adults with MCI. Cerebral blood-flow/oxygen measures are surrogates. Frequent manufacturer involvement, small samples and formulation differences limit certainty.
    PMID 34415830 PMID 36807425 PMID 26782228 PMID 27526146 PMID 38908296
  • Mitochondrial biogenesis signaling Limited · benefit
PGC-1alpha/SIRT1/CREB/AMPK mechanisms are mostly cell/animal evidence. Small human metabolite/signaling changes do not establish larger functional mitochondrial capacity.
    PMID 29185343 PMID 19861415 PMID 32860006 PMID 24231099
  • Inflammatory markers Limited · benefit
A 10-person human metabolic study (5 women, 5 men), 0.3 mg/kg/day over about 76 hours, reported CRP/IL-6 reductions. The small design lacked a parallel placebo group; not treatment evidence for inflammatory disease.
    PMID 24231099
  • Aerobic exercise performance Limited · no benefit
In 23 untrained men, 20 mg/day for 6 weeks with endurance training did not improve aerobic performance versus placebo, despite a PGC-1alpha marker increase.
    PMID 31860387
  • Lipids Limited · no benefit
After 12 weeks at 20 mg/day, triglycerides were unchanged. A marginal LDL reduction occurred only in the higher-baseline-LDL subgroup (at least 140 mg/dL; 11/29 participants), not a general lipid benefit.
    PMID 26226960
  • AMH and fertility No established evidence
Single-arm open-label study of 50 healthy women aged 25-42: the catalog title reports no overall AMH change and exploratory subgroup findings. Without controls, it cannot establish improved ovarian reserve, fertility or a causal benefit.
    PMID 42500134
  • Sleep, mood and subjective fatigue No established evidence
The frequently cited sleep/stress/fatigue report is not indexed in the supplied catalog; no indexed RCT basis or numerical effect is supplied. Anecdotal immediate energy does not establish a clinical benefit.

Reader questions

Should I use 10-20 mg or 50-100 mg?

Ten to twenty mg/day is the main supplied human trial range. Higher-dose internet regimens are not validated here, and no UL is established. Trial ranges are not personal dosing advice.

Is PQQ neurotoxic?

This dossier does not establish neurotoxicity at studied human doses. It also cannot establish long-term safety: rodent/high-dose reports are not a human threshold, and the alarming article cited by readers has unresolved provenance. Do not solve uncertainty by increasing the dose.

Is BioPQQ better than generic disodium PQQ?

The named salt is the same chemical form. Identity, purity and independent testing matter; no supplied human trial proves a branded salt more effective than an equivalently verified generic.

Does PQQ improve fertility?

The supplied uncontrolled AMH study had no overall change and exploratory subgroup findings. It does not establish pregnancy or fertility benefits; decisions belong with the treating fertility clinician.

Stack interactions

  • Caution CoQ10/ubiquinol and ALCAR [STACK // UNPROVEN_SYNERGY]
Popular mitochondrial stacks have mechanistic rationales but no established human synergy in the supplied dossier.
  • Caution Atorvastatin [PRECLINICAL // NOT_HUMAN_DDI]
Combined mitochondrial/inflammatory effects were studied in obese rats, not a demonstrated human drug interaction.
    PMID 32535101
  • Caution Diabetes medication [CAUTION // CLINICAL_REVIEW]
Potential additive metabolic effects are theoretical; no documented clinical interaction is supplied.
  • Caution Cancer therapy [CAUTION // CLINICAL_REVIEW]
Antioxidant-related concerns are theoretical and nonspecific to PQQ here; changes during chemotherapy/radiotherapy require the treating oncology team.

Red flags: interactions and who should avoid it

Interactions

No well-documented human drug-interaction study is supplied. Diabetes-drug cautions are theoretical; atorvastatin combination findings are from rats. Oncology-treatment concerns are nonspecific antioxidant theory and require the treating team, not a home protocol. CoQ10/ubiquinol and ALCAR combinations have no established human synergy beyond component effects.

Avoid or ask a doctor first if

Avoid self-directed use during pregnancy, breastfeeding, childhood or fertility treatment because human safety data are inadequate. Review chronic disease/polypharmacy before use. Short RCTs reported reasonable tolerability at 10-20 mg/day; GI discomfort and stimulation/insomnia reports are anecdotal. High-dose rodent renal/toxicity signals do not define a human safe dose, and the cited toxicity article lacks resolved provenance in this dossier.
